Enterobacteria phage G4 is a bacteriophage capable of infecting susceptible bacterial cells. The phage was originally isolated from samples of raw sewage and infects E. coli.[1] Its isometric capsid contains a single-stranded circular genome of about 5.5 kbp. Phage G4 bears high genetic and structural similarity to the phage ΦX174, though the two phage genomes are up to 39 percent different by sequence.[2]

Full-sized genomes of bacteriophage G4 molecules were completed from short overlapping synthetic oligonucleotides by direct assembly polymerase chain reaction and ligase chain reaction followed by fusion polymerase chain reaction with flanking primers.

The synthetic phage G4 genomes, after transformation into E. coli, were translated by the host cells and led to complete and infective G4 viral particles.[3]
