The Memphis, Clarksville and Louisville Railroad received its charter on January 28, 1852, from the state of Tennessee. The 83-mile (134 km) line of 5 ft (1,524 mm)[1] gauge connected with the Memphis and Ohio Railroad and the Louisville and Nashville Railroad (L&N) connects between Memphis and Louisville.

The railroad had a major strike in 1868 and closed for several days. After the strike, the L&N took control of the Memphis, Clarksville and Louisville Railroad.
